Intended Use
Human insulin-like growth factors binding protein 1 (IGFBP-1) PROM Test is a visually, interpreted, qualitative immunochromatographic test for detection of IGFBP-1 (insulin-like growth factor-binding protein 1) in vaginal secretions during pregnancy, which is a major protein marker of the amniotic fluid in a vaginal sample. Human insulin-like growth factors binding protein 1 (IGFBP-1) PROM Test is intended for professional use to help diagnose the rupture of fetal membranes (PROM) in pregnant women.
Introduction
Premature rupture of membranes (PROM) is a condition that can occur in pregnancy. And it affects 5 to 10% of pregnancies and is associated with serious complications, including premature delivery, Fetal Distress, puerperal endometritis and neonatal sepsis. 25%-35% premature delivery are caused by PROM. insulin-like growth factor-binding protein 1(IGFBP-1) is one of the main protein in amnioticfluid from Pregnancy 3 months to delivery. And the concentration of the IGFBP-1 in amnioticfluid is 100-1000 times to it in serum. When the membranes rupture, the IGFBP-1 enter into cervix and vagina, so it can be detected in vaginal secretion.
Principle
The PROM Rapid Test Device has been designed to detect IGFBP-1 in vaginal secretion through visual interpretation of color development in the internal strip. The membrane was immobilized with IGFBP-1 antibody on the test region and the conjugate pad was pre-coated with colored IGFBP-1 antibody colloidal gold conjugates. After specimens were added, the gold-conjugates move along the membrane chromatographically by capillary action and antibodies get to the test region. If sufficient IGFBP-1 is present in the sample, it will react with colored IGFBP-1 antibody colloidal gold conjugates in conjugate pad, the mixture then moves to the membrane, and combine with IGFBP-1 antibody on the test region. As a result, a colored band will form at the test region of the membrane.
If no IGFBP-1 is present in the sample, colored IGFBP-1 antibody colloidal gold conjugates will not be captured by IGFBP-1 antibody on the test region, so there is no colored band appearing at test region.
Therefore, the colored band on the test region indicates a positive result. And appearance of a colored band at the control region serves as a procedural control. This indicates that proper volume of specimen has been added and membrane wicking has occurred.
